Description
Für die Forschungsgruppe "Redox and Metalloproteine" am MPI für Biophysik, die an der Entwicklung von Methoden zur elementaren Identifizierung in dosisempfindlichen biologischen Proben mittels Elektronenmikroskopie arbeitet, wird ein Panther STEM-Upgrade sowie ein Win10 Upgrade für zwei bereits am Institut bestehende Elektronenmikroskope beschafft. Die analytischen STEM-Techniken, einschließlich der STEM-Elektronen-Energieverlustspektroskopie (EELS) und des winkelaufgelösten 4D-STEM, liefern wichtige Informationen über die Lage bestimmter Elemente innerhalb einer Probe im Elektronenmikroskop. Diese Techniken erfordern jedoch eine hohe Gesamtelektronendosis und werden in der Regel nur bei dosisverträglichen Proben eingesetzt. Die von der Gruppe Redox und Metalloproteine am MPI für Biophysik geleiteten Arbeiten zielen darauf ab, diese Techniken für die Anwendung auf dosisempfindliche kryokonservierte biologische Proben zu übertragen (Pfeil-Gardiner et al. Elemental mapping in single-particle reconstructions by Reconstructed Electron Energy-Loss (REEL) analysis, Nat. Methods 2024).
